    Overview


    The development of CRISPR/Cas9 over the past decade has enabled the development of precise genetic and stem cell therapies for treating many diseases.

    However, there are several hurdles in applying this technology to treat diseases affecting the lungs.

    The Vaidyanathan laboratory is investigating the use of CRISPR/Cas9 based genome editing to treat diseases affecting the lungs, starting with cystic fibrosis (CF).

    The focus of this project is to develop an airway stem cell therapy for treating cystic fibrosis (CF). This would be a great opportunity to gain experience in genome editing, stem cell biology and tissue engineering. Our lab is in the Center for Gene Therapy at Nationwide Children's Hospital (NCH).

    The NCH Center for Gene Therapy has a long track record of training leaders in gene therapy, has been the site of the development of the first FDA-approved AAV-mediated gene therapy for neuromuscular disease (SMA).

    Our lab is looking for a Research Assistant to join our energetic and innovative team to carry this mission forward.

    The individual will work full-time, under the guidance of the PI, Dr. Sriram Vaidyanathan.

    The position will involve molecular biology, cell culture, working with mice (after sufficient training) and some lab support tasks such as ordering supplies and equipment, maintaining lab records, and adhering to safety regulations .

    The individual will perform experiments supporting isolation of cells from airway tissue, molecular biology, cell culture of airway stem cells, differentiation of airway stem cells in air-liquid interface and animal work.

    They may be expected to troubleshoot existing protocols, develop novel protocols and provide technical and scientific advice to other research personnel in lab.

    The individual will acquire other career development opportunities, such as presenting at lab meetings, participating in journal clubs, and the possibility of authorship on publications that will arise from their work.

    Responsibilities


    PRINCIPAL DUTIES AND RESPONSIBILITIES

    • Perform complex (lab and/or field) experiments for a major portion of the research project independently or semi-independently in accordance with protocols approved by the principal investigator. Such experiments may involve the use of laboratory animals or human subjects any may require evening and/or weekend duties.
    • Prepares and tests substrate, conducts assays, and prepares tissue cultures for use in research studies, as applicable. Collects blood, urine, and other samples as needed. Such activities may be performed independently and may require evening and weekend hours.
    • Ensures accuracy of laboratory analytical procedures as required by specific research projects.
    • Collects and controls experimental data and patient information; and maintains accurate daily logs of experiments including methods, results, and conclusions.
    • Interprets the results of data analyses with input from the principal investigator.
    • Perform ongoing literature review to remain current with new procedures and related research; present findings for lab work group.
    • Assists the principal investigator in the preparation of scientific publications and grant proposals. May present ongoing work and findings to colleagues at academic conferences.
    • Make recommendations about refinement and/or replacement of laboratory analytical procedures.
    Implements such refinements or new procedures with input from the principal investigator.

    • Orders supplies needed to conduct research; maintains inventory of equipment and supplies; and assures that research equipment is in good working order.
    • Collaborates in the preparation of research budgets, and monitors expenditure of funds to assure that studies are conducted within budgetary constraints.
    • Instructs other laboratory workers and students in standard methods, practices, procedures, and equipment operations.
    • Maintains a clean working area and performs general laboratory maintenance. Adheres to proper safety techniques and procedures.
    • Performs miscellaneous related duties.
    Qualifications

    KNOWLEDGE , SKILLS AND ABILITIES REQUIRED


    • Bachelor's Degree in a relevant field of science from an accredited college.
    • Six months to two years of relevant laboratory experience. Coursework in anatomy, physiology, statistics, relevant research methods and radiation desirable.

    Percent of time spent on:

    • 50% Preparing test substrates, conducting assays, preparing cultures
    • 25% Interpreting results of data analyses with PI
    • 11% Miscellaneous paperwork associated with research
    • 10% Ordering supplies
    • 02% Preparation of budget
    • 02% Instructing other lab personnel

    Minimum physical requirements:

    • 100% Kneeling / reaching
    • 100% Finger dexterity
    • 100% Talking / hearing / seeing
    • 33% Lifting / carrying weight 0-20 pounds for 1-5 minutes
