    This position will be supporting both the Anderson Lab at Brigham and Women's Hospital as well as the Transcriptomics Platform of the Gene Lay Institute of Immunology and Inflammation.

    In the Anderson lab, this position will work under minimal supervision to execute research experiments involving a variety of wet-lab techniques including genomics e.g.

    single-cell and bulk RNA-seq.

    They will assist other laboratory members to execute experiments, as well as perform some maintenance tasks to benefit the lab as a whole.

    They will be responsible for collecting and analyzing their own data as well as presenting their work to the lab and their supervisor.

    At the Transcriptomics Platform, this position will work closely with the Platform manager to assist in running samples through an advanced robotics pipeline that creates sequence-ready NGS libraries for both bulk and single-cell RNA sequencing.

    This position will help with sample preparation for downstream genomics applications along with assisting the Platform manager in general up-keep of the platform.


    PRINCIPAL DUTIES AND RESPONSIBILITIES:

    • Under general supervision, will breed and maintain common use mouse strains and use molecular (PCR) and cellular (flow cytometry) techniques to genotype transgenic and knockout mice.
    • Maintain records of mouse breeding using the Transnetyx Colony Management system.
    • Perform basic cell culture and in vitro assays and analyze by multi-parameter flow cytometry.
    • Perform DNA quantification, plasmid DNA isolation, and DNA analysis by restriction digest.
    • Perform in vivo experiments including tumor implantation, measurement, and treatment of tumor-bearing mice, as well as induction and monitoring of autoimmune disease.
    • Perform gene expression analyses using quantitative PCR.
    • Perform general lab management duties for both the Anderson Lab and the Transcriptomics Platform, including ordering supplies, maintaining reagent inventory, and assisting with the orientation of new staff.
    • Coordinate daily schedule of experiments.

    Responsible for experimental data management:
    collect, record, tabulate, and present data. Create graphs of experimental data, perform statistical tests, and discuss progress of work regularly with supervisor.


    • Coordinate with medical staff to collect patient samples and process these samples to obtain single cell suspensions for downstream NGS library preparation.
    • Work with the Transcriptomics Platform manager to prepare both bulk and single cell RNA sequencing libraries
    • All other duties as assigned.

    QUALIFICATIONS:
    BS required.

    New graduate with relevant course or project work.


    SKILLS/ABILITIES/COMPETENCIES REQUIRED:

    • High degree of computer literacy
    • Experience working with mice is desired
    • Sound analytical and organizational skills.
    • Requires good oral and written communication skills.
    • Must be able to logically and effectively structure tasks and set priorities
    • Ability to identify potential problems and trouble-shoot solutions.
    • Strong interpersonal communication skills.

    WORKING CONDITIONS:
    Normal laboratory environment.

    Radioactive and hazardous materials are used in some of the experiments conducted.

    Animal and human tissue will be used.
