    Scribe Therapeutics is a molecular engineering company focused on creating best-in-class in vivo therapies that permanently treat the underlying cause of disease.     We are seeking a Scientist with experience in RNA biochemistry and an adaptable skillset of computational, biophysical, and biochemical techniques to probe the structure/function of single-guide RNAs (sgRNAs). The candidate will join the Process Science analytical team to study RNA folding computationally and experimentally for its CRISPR by DesignTM platform. The ideal candidate should have a good understanding and knowledge of RNA biochemistry and methods for probing RNA structural ensembles. We are seeking experienced, enthusiastic, creative, and highly motivated individuals who are comfortable working in a fast-paced entrepreneurial CMC environment.

    Key Responsibilities:

    • Structure probing of development candidate sgRNAs using enzymatic or chemical structure mapping techniques such as DMS MaP-seq and/or high-throughput SHAPE
    • NGS library design, preparation, QC, and pipeline construction for NGS
    • In-silico RNA structural modeling and alternative RNA structure ensemble deconvolution
    • Construction of cloud-based computational environments for RNA structure analysis
    • sgRNA sequencing for identity confirmation by NGS
    • Biophysical studies of sgRNA binding affinity for CasX using Biolayer Interferometry (BLI)
    • Biophysical studies of DNA target-binding affinity of CRISPR-CasX effector complexes using BLI
    • Functional characterization (in vitro DNA cleavage) of differential CRISPR-Cas RNP complexes by electrophoretic mobility shift assay (EMSA) or anion exchange (AEX) chromatography
    • Work collaboratively in a multi-disciplinary environment with Process Science and Research teams
    • Maintain detailed records of experimental methods and data in electronic laboratory notebook (Benching)
    Required Skills and Background:
    • PhD with 0-2 years of experience, or MS with 6+ years of experience in an academic/ industry setting. Degree in Chemistry, Biochemistry, Chemical Engineering, Bioengineering, Pharmaceutical Sciences, or related discipline
    • Excellent oral and written communication skills
    • Effective organizational and time management skills
    • Strong focus and attention to detail
    • Ability to work both independently and collaboratively in a fast-paced, interdisciplinary research team
    Preferred Skills and Background:
    • Familiarity with CRISPR technologies and therapeutic approaches
    • In silico RNA folding / prediction algorithms
    • RNA biochemistry
    • Experience in NGS based assay development and implementation
    • Hands-on experience with Illumina, PacBio, and/or Nanopore sequencing platforms
    • Bioinformatic pipelines; Python, MATLAB

    At the time of posting, the base pay wage range for this role is $100,000-$130,000 per year. The offered pay range will depend on internal equity and the candidate's relevant skills, experience, qualifications, training, and market data. Additional incentives are provided as part of the complete package in addition to comprehensive medical and other benefits.
